Multilocus sequence typing (MLST) provides a new approach to molecular epidemiology that can identify and track the global spread of virulent or antibiotic-resistant isolates of bacterial pathogens using the Internet. MLST databases, together with interrogation software, are available for Neisseria meningitidis and Streptococcus pneumoniae and databases for Streptococcus pyogenes and Staphylococcus aureus will be released shortly.  相似文献

In some ecosystems a small suite of species can determine community‐level patterns of species richness by acting as either ‘accumulators’ or ‘repellers’; that is, the richness of the immediate neighbourhood of such species departs from that expected on the basis of a given null model. Using the individual species‐area approach, we evaluated community‐level spatial pattern in four species‐rich shrublands (two 40 × 40 m and two 30 × 30 m plots) by assessing the frequency of accumulator and repeller species and whether any such species were associated with specific life‐history characteristics. Few species departed from the expectations of the null spatial model that we used, although, at three of the four sites, accumulators were more common than repellers. Departures from the null model we assessed were most prevalent within just 1 m of focal individuals and were not consistently associated with specific life‐history traits. Model‐based clustering suggests that there are distinct sub‐communities in each of the four communities, but while internally spatially cohesive, these sub‐communities intermingle and their membership is not predictable from the life‐history traits of their constituent species. Comparable analyses in other species‐rich systems have also detected a similar absence of spatial interactions. The disturbance regime in the shrubland communities we consider is markedly different from those in the forest ecosystems where previous studies have been concentrated (recurrent fire versus infrequent gap‐phase dynamics) and resources are more limiting. Thus, our results provide further support for the generality of a model of stochastic geometry, likely underpinned by stochastic dilution effects, in species‐rich plant communities.  相似文献

This study examined the onset of puberty, follicular dynamics, reproductive hormone profiles, and ability to maintain pregnancy in cloned heifers produced by somatic cell nuclear transfer. Four adult somatic cell-cloned heifers, derived from a 13-yr-old Holstein cow, were compared to 4 individual age- and weight-matched heifers produced by artificial insemination (AI). From 7 to 9 mo of age, jugular venous blood samples were collected twice weekly, and from 10 to 11 or 12 mo of age, blood sampling was carried out every other day. After the heifers reached puberty (defined as the first of 3 consecutive blood samples with peripheral plasma progesterone concentrations of >1 ng/ml), ultrasound examination of ovaries and jugular plasma sample collection were carried out daily for 1 estrous cycle. Cloned heifers reached puberty later than controls (mean +/- SEM, 314.7 +/- 9.6 vs. 272 +/- 4.4 days and 336.7 +/- 13 vs. 302.8 +/- 4.5 kg for clones and controls, respectively; P < 0.05). However, cloned and control heifers were not different in estrous cycle length, ovulatory follicle diameter, number of follicular waves, or profiles of hormonal changes (LH, FSH, estradiol, and progesterone). Three of the 4 clones and all 4 control heifers became pregnant after AI. These results demonstrate that clones from an aged adult have normal reproductive development.  相似文献

3H-l-Phenylalanine is incorporated into a range of phenylpropanoid compounds when fed to tobacco cell cultures. A significant proportion of (3)H-trans-cinnamic acid formed from (3)H-l-phenylalanine did not equilibrate with exogenous trans-cinnamic acid and therefore may be rapidly channeled through the cinnamate 4-hydroxylase (C4H) reaction to 4-coumaric acid. Such compartmentalization of trans-cinnamic acid was not observed after elicitation or in cell cultures constitutively expressing a bean phenylalanine ammonia-lyase (PAL) transgene. Channeling between PAL and C4H was confirmed in vitro in isolated microsomes from tobacco stems or cell suspension cultures. This channeling was strongly reduced in microsomes from stems or cell cultures of transgenic PAL-overexpressing plants or after elicitation of wild-type cell cultures. Protein gel blot analysis showed that tobacco PAL1 and bean PAL were localized in both soluble and microsomal fractions, whereas tobacco PAL2 was found only in the soluble fraction. We propose that metabolic channeling of trans-cinnamic acid requires the close association of specific forms of PAL with C4H on microsomal membranes.  相似文献

The ability of gonadotropin‐releasing hormone (GnRH) immunization to disrupt estrous cycles in captive white‐tailed deer (Odocoileus virginianus) was tested. Four does were each injected subcutaneously with 1 mg of a GnRH analog‐ovalbumin conjugate, using a diethylaminoethyl (DEAE)‐dextran solution as the adjuvant. Control deer (n = 4) received ovalbumin alone in DEAE‐dextran. The immunization schedule consisted of a primary immunization on 30 December 1994, followed by three booster immunizations at 4‐week intervals. In addition, a booster was administered the following year (3 Nov 1995) before the start of the breeding season. Serum from control females did not contain GnRH antibodies, and estrous cycles of these deer were not disrupted (as determined by serum progesterone concentrations). In contrast, all GnRH‐immunized deer had detectable GnRH antibody titers by 1 week after the first booster. Estrous cycles were disrupted in two of these four deer. The booster given the following year failed to stimulate an increase in mean antibody titers, and all deer began to cycle, including one that had maintained high titers from the previous year. These data suggest that a GnRH analog conjugated to ovalbumin, using DEAE‐dextran as adjuvant, is immunogenic in female white‐tailed deer and may be able to prevent ovulation in some individuals. However, pending further work to increase the immunogenic and biological responses and to decrease the variation among animals, this vaccine does not appear to be an effective means of contraception for deer. Three roselle, Hibiscus sabdariffa L. varieties (Sudani, Masri and White) were cultivated at three different locations to recognize the transportation ability of roselle cultivation from the narrow old valley land to broad new land in Egypt. Qena as origin in situ old land, El-Kanater as ex situ old land and Nubaria as ex situ new land were the considered locations. Six growth quantitative characters and bolls infestation by spiny bollworm, Earias insulana were evaluated. Growth characters of roselle plants were affected significantly by either variety or location. Qena region was more suitable for roselle plant growth as judged with plant height, number of branches, number of fruits and sepals dry weight, followed by Nubaria followed by El-Kanater. Whereas, plants grown at Nubaria produced more fresh sepals weight than Qena or El-Kanater grown plants. As for Sudani, Nubaria exhibited the tallest plants, with the highest number of fruits and the heaviest fresh sepals as compared with the corresponding plants in Qena or El-Kanater. Values of broad sense heritability were highest for all characters in Qena. While the number of fruits per plant had the highest heritability in all locations. Dry sepals yield had highly significant correlation with all studied characters except percentage of water loss in Qena and Nubaria. Path coefficient analysis confirmed that fresh sepals yield had the highest direct and indirect effects on dried sepals yield. Chemical constituents responsible to sepal quality tended to produce significant variations due to the changes in varieties or locations. The highest levels of anthocyanins and sugars were achieved by Sudani variety, but the highest levels of free amino acids and total soluble solids were recorded for Masri variety. Moreover, Nubaria region was the most favourable for the accumulation of more anthocyanins in the sepals of all varieties followed by Qena. Plants grown at Qena produced sepals with the highest levels of sugars, free amino acids, organic acids and total soluble solids, followed by Nubaria followed by El-Kanater plants. Infestation with spiny bollworm Earias insulana was increased from Sudani up to Masri up to White varieties. Plants grown at Nubaria had the lowest number of attacks by bolls in all varieties, followed by those at El-Kanater followed by Qena plants. Spiny bollworm infestation was positively correlated with the number of branches and dry sepals weight, but negatively correlated with sepal moisture loss and anthocyanin contents. These findings clearly indicated that the Nubaria region was considered as a promising reclaimed area suitable for roselle cultivation, especially for Sudani, the most economic variety.  相似文献

Plant community composition and its likely environmental controls were investigated for 200 sample plots (each 100 m2) from Mediterranean-type vegetation in the Little Desert National Park, Victoria. TWINSPAN classification revealed four readily identifiable vegetation types; mallee-broombush, heathland, stringybark open woodland, and an assemblage intermediate between mallee-broombush and heathland referred to here as broom-heath. Mallee-broombush was found on Parilla Sands characterized by high Ca levels relative to heathland and stringybark open woodlands on unconsolidated Lowan Sands. The first axis of a 2 dimensional non-metric MDS ordination also divided heathlands (high axis scores) from mallee-broombush (low scores), while the second separated these vegetation types from stringybark woodlands and broom-heath. Vector-fitting revealed significant correlations between the locations of samples in ordination space and exchangeable soil Ca, soil colour, aspect and Shannon–Weiner diversity. Highest species richness/diversity was associated with the ecotonal area between Parilla and Lowan Sands (i.e. broom-heath) where a number of species characteristic of different assemblages had overlapping ranges. The fire-sensitive conifer, Callitris rhomboidea, was preferentially located in stringybark woodland and broom-heath vegetation types. Its presence was positively associated with high species richness and aspects having a southerly component. Four Callitris stands sampled for population structure were all > 40 years old and showed evidence of interfire recruitment from seeds released by old, serotinous cones. Overall, results suggest that variations in plant community composition and structure in the eastern block of the Little Desert are primarily due to variations in soil properties associated with the distribution of the two dominant substrate types, Parilla Sand and Lowan Sand. However, the interplay of topography and fire behaviour has probably been more important than substrate type in determining the distribution and population structure of longer-lived, fire-sensitive species such as Callitris rhomboidea.  相似文献
